My system is to support up to 1000 agents. I don't think using Asterisk is the best choice at the moment.
Kamailio is a SIP proxy and registrar, not a PBX neither a SIP voice/media application server. You can use Kamailio to do load balancing and failover in front of various Asterisk/FreeSwitch/other_media_server, but Kamailio itself cannot manage "agents", "queue" and so (it's a SIP proxy). Kamailio is not a "cool Asterisk".
