I want to use OpenSER + mediaproxy with TLS/SRTP.
Is mediaproxy able to handle the srtp stream?

But now I have a next question: Has anybody done conferences (+ mailboxes) with TLS/SRTP? I know that there is a TLS and a SRTP fork of Asterisk which manages this and Freeswitch should also be able to handle TLS/SRTP.
So my question: Has anybody managed to run secure phone conferences with the Asterisk fork or Freeswitch, or is there a different way of doing this?

I never tried it but as SRTP is the same as RTP (from mediaproxies viewpoint) it shouldn't be a problem.

if you want to/can use ZRTP for SRTP key agreement, you can check out SEMS (http://iptel.org/sems) which con be compiled with ZFone SDK for ZRTP support.

I have seen that, but this and requires a softphone and additional software for each client. I would like to use it with a Snom 370 or Polycom 650 SIP phone.

If you don't find what you are looking for in Freeswitch or Asterisk, you might try to use zfone's SRTP API directly as it might not need big modifications to sems' core (see this doc http://zfoneproject.org/docs/sdk/man/html/guide.html#srtp_usage ).
