For some reason it doesn't meet my requirement.
Am able to change request uri with To header values as follows:
avp_pushto("$ruri/username", "$tU");
avp_pushto("$ruri/domain","$td");
avp_delete("*");
Then doing a lookup("location")
It is working fine.

From the example I understood you want to replace request uri with To
