Finally as you suggested in the answer we do change DB and then force a reload of the status of the nodes through the ds_reload MI command (http://kamailio.org/docs/modules/stable/modules_k/dispatcher.html#id2513000)  and it works great

I say this because I would like to update active and inactive nodes from a 3rd party application modifying the flags column but I need to know also how many time will it take for the change to be applied.
the list of addresses is loaded at startup. To reload it, you have to send a MI/RPC command (e.g., 'kamctl fifo ds_reload', or 'sercmd dispatcher.reload', or via xmlrpc). If you just want to change the flags, there is a mi/rpc command for it:
